Fresh liver tissue sections (300 μm thick) were made from surgically resected fibrotic livers from liver disease patients using Krumdieck Tissue Slicer MD6000 (Alabama Research and Development) (10). The tissues were cultured with captopril (100 μM), erlotinib (5 μM), or DMSO vehicle control for 48 hours and harvested for gene expression analysis as described above. For ex vivo tissue culture, we used a higher concentration compared with culture of cell lines according to ref. 65.
