At the end of the study (17 weeks post HFD), all mice were euthanized by CO2 asphyxiation followed by cardiocentesis. Tissues were collected under yellow lights to prevent degradation of retinoids. Sera were separated and stored at −80°C until triglyceride and retinoid analyses. Liver, lung, brain, testes, and adipose depots (epididymal, retroperitoneal, mesenteric, inguinal, and brown adipose tissue) were dissected, weighed and small pieces were fixed in 10% phosphate-buffered formalin or snap-frozen in liquid nitrogen and stored at −80°C.
