To induce gametogenesis, cells were scraped from a week-old plate and resuspended in N-free TAP to a final concentration of 5 × 107 cells mL−1. After incubation for 3–5 h in the light, equal numbers of plus and minus gametes were mixed and allowed to mate. To quantify gametic activity, the mating efficiency was calculated at 1.5 h after mating based on the percentage of gametes engaged in the formation of quadriflagellate cells.
