Briefly, 25 μl of 6% phenol and 50 μl of samples were added and mixed in the optically clear flat-bottom 96-well plate (Costar 3599, Corning, Corning, NY, United States), and then 125 μl of concentrated sulfuric acid was added directly into the solution. The mixture was blown with a pipette, and allowed to stand for 30 min at room temperature. The optical density (OD) was measured at 490 nm by Multiskan GO 1510 (Thermo Fisher, Vantaa, Finland). The OD490 indicated the yield of polysaccharides.
