Mouse Achilles tendons and mouse 3D tendon-like constructs were fixed in 4% paraformaldehyde for 12 h at 4°C, and after several washes in phosphate-buffered saline (PBS) and cryo-preservation in 30% sucrose in PBS were embedded in cryomedium (Surgipath Cryogel®, Leica Microsystems, Vienna, Austria). Subsequently, 12 µm cryosections were prepared using a Leica CM1950 cryostat.
