The protein expression of p-JAK1, 2, 3 (Rabbit mAb; CST), p-STAT1, 2, 3 (Rabbit mAb; CST), CDK2 (Rabbit Polyclonal; Proteintech), cyclin E (Rabbit Polyclonal; Proteintech), p21 (Rabbit Polyclonal; Proteintech), VPS25 (Rabbit Polyclonal; Proteintech), METTL3, METTL14 and YTHDC1 (Rabbit Polyclonal; Proteintech) was detected by western blot as described previously [23, 24]. Briefly, cells were lysed by Sample Buffer, Laemmli 2 × Concentrate (Sigma, MO, USA) via the manufacturer's protocol. Next, the proteins were isolated by 10% SDS–PAGE gel and transferred to nitrocellulose membranes. After immunoblotting with primary and secondary antibodies, the protein bands were incubated with enhanced chemiluminescence (ECL) and visualized using a Tanon 5200 Chemiluminescence Imaging System (Shanghai, China).
