S. aureus strains were grown to midlog phase (OD600 ∼0.4), washed by centrifugation, and resuspended in 0.1% volume/volume Triton X-100 in phosphate-buffered saline to a final OD600 of ∼0.8, with or without addition of antibiotics. Cell lysis was monitored by OD600 measurements.
