As previously published (30), Vero 76 cells were seeded in 96-well plates and grown overnight (37°C) to confluence. A sample of the supernatant fluid from each compound concentration was collected on day 3 (day 4 for POWV and day 7 for USUV) postinfection (3 wells pooled) and tested for virus titer using a standard endpoint dilution CCID50 assay and titer calculations using the Reed-Muench equation (31). The concentration of compound required to reduce virus yield by 90% (EC90) was determined by regression analysis.
