Broth macrodilution was performed with arthroconidia in tubes for MIC and MFC determinations as previously described (20, 22–24). In brief, the initial inoculum was 103 arthroconidia/ml with a 2-fold dilution range, starting at 100 μg/ml for FCZ and 20 μg/ml for nikZ, and was incubated at a temperature to approximate human body temperature (35°C to 37°C). The MIC value was determined when the control growth was 4+, and the endpoint was defined as the first tube without growth. MFC was determined by subculturing the isolate on agar and defined as killing ≥96% of the initial inoculum. Both readings occurred after 7 days of incubation.
