After 21 days of osteogenic differentiation, ALP staining was applied to assess the osteoblastic mineralization of BMSCs. The ALP activity of cell lysates was then detected by spectrophotometric measurement of p-nitrophenol release using an enzymatic colorimetric ALP kit (Roche, Switzerland). The mineralized nodules were quantified by a microplate reader at 420 nm.
