Cells were incubated and treated with 0 or 30 μM MnTE-2-PyP overnight. Cells were trypsinized, washed, counted and re-suspended in media without FBS. The serum-starved cells were re-plated on a PET track-edged membrane, pore size 8.0 μm (Fisher Scientific, Pittsburg, PA, USA) set inside the well of a six well plate at a concentration of 1.0 × 106 cells/well. The membrane was situated between a top chamber containing media without serum and a bottom chamber filled with serum media. Cells were allowed to migrate across the membrane for 48 hours. The cells and media were isolated from the top and bottom chambers and enumerated.
