HEK293T cells were transfected with titrating amounts of FLAG-HBZ, FLAG–APH-2, or the control expression vector (approximately 2,000 ng total DNA per well) in a 6-well dish using the Lipofectamine 2000 transfection reagent (Life Technologies) according to the manufacturer's instructions. When indicated, 200 ng of the p65 and IRF-1 expression vectors was used. Immunoblot analysis was performed 48 h after transfection to compare the levels of transfected proteins.
