The MTT method was used to assess cell viability. Cells were seeded at a concentration of 5×103 cells per well in 96-well plates. After growth for 24 h, cells were treated with the indicated doses of rhHMGB1 for 24, 48 and 72 h. Cell viability was measured using the MTT (3-[4,5-dimethyl-2-thiazolyl]-2,5- diphenyl-2H-tetrazolium bromide) assay kit (Sigma) and absorbance was read using a microplate reader (Bio-Rad) at 492 nm. All experiments were carried out in triplicate.
