After DPCs were grown overnight to form a confluent monolayer, pAdTrack-CMV-TCF4 and pAdeno-MCMV-MAD2B were transduced into DPCs, individually or in combination, at a multiplicity of infection (MOI) of 50. plRES2-TCF4, plRES2-MAD2, and siMAD2B, a vector expressing short hairpin RNA against MAD2B that was constructed previously22, were transfected, individually or in combination, using Lipofectamine 2000 (Invitrogen), following the manufacturer’s instructions. The empty vector was used as a negative control.
