Intracellular calcium concentration ([Ca2+]i) was assessed in freshly isolated intact islets using a dual wavelength PTI system (PTI, Monmouth, USA) fitted on an inverted Zeiss microscope allowing ratiometric measurements using the probe fura-2AM (Invitrogen, Paisley, UK) as described previously (15).
