POD activity was quantified in a spectrophotometer by oxidizing the 0.1 mL protein sample in 3 mL of 0.05 M phosphoric acid buffer, pH 5.5, 2 mL of 2% hydrogen peroxide (H2O2) as the oxidant, and 36 μL guaiacol as the hydrogen donor per 50 mL buffer. The oxidation of guaiacol was measured at 470 nm at 25 °C and was expressed in U g−1 FW min−159.
Enzyme activity was defined as “an increase in absorbance of 1 unit enzyme at 470 nm min−1 at 25 °C”. The specific activity of POD was expressed in U mg protein−1.
