Several passivation protocols were evaluated to optimize the quality of the passivation layer on PDMS substrates as well as the quality of the patterned protein after protein incubation (Table 1). 3-Aminopropyltriethoxysilane (APTES; A3648, Sigma-Aldrich) and poly-l-lysine (PLL, P4707, Sigma-Aldrich) were used in combination with mPEG-succinimidyl valerate (mPEG-SVA; MW 5000 Da, Laysan Bio) as well as Pluronic F-127 (P2443, Sigma-Aldrich) to passivate the PDMS substrates in various ways.
