The activity of the mitochondrial enzyme citrate synthase was quantified by measuring the formation of thionitrobenzoate from 5,5-dithiobis-2-nitrobenzoate after the addition of 0.3 mM acetyl-coenzyme A and 0.5 mM oxaloacetate. The production on thionitrobenzoate was measured spectrophotometrically at 412 nm for 2 min.
