The cell counter (CASY cell counter) parameters were set as described in Table . We chose the size range from 27.75 to 45 μm as pollen particle because this range covered pollen peak from all samples without contamination from small/large particles. A 200-μL pollen suspension was mixed with 10 mL of CASYton (OMNI Life Science). Particle numbers were counted using a cell counter by sampling 400 μL three times. Viable cells were calculated as the total pollen number per strobilus using the following equation:
Taken together, cell counter counted 1/42.5 of pollen grains of one strobilus.
