Gene edition by CRISPR/Cas9 technology

After 293T, THP‐1 cells, and PBMCs were seeded, medium was replaced with DMEM containing polybrene (8 μg/ml) (Sigma) and lentiviruses expressing Cas9 and specific sgRNAs for 18 h. Cells were selected for puromycin resistance, and polyclonal pools of transduced cells were used for clonal selection and following experiments.
