MIN6 cells (2×10^5 cells well-1) or isolated mouse islets (15 islets well-1) were seeded in 48-well plates，MIN6 cells or the islets were pre-incubated overnight in KRBH balanced buffer containing 0.2% BSA supplemented with 2.5 mmol/l glucose, and were incubated for 2 h in 2.5 mmol/l or 33.3 mmol/l glucose.
