Equipment

pH试纸;600ml、1500ml烧杯或烧瓶;量筒;搅拌器。

Procedure

  1. 配制EDTA储存液(以500ml,0.5M EDTA为例)

    1. 称取93.05gEDTA二钠盐

    2. 溶解在400ml去离子水中,用1M NaOH溶液调节pH至8.0使EDTA完全溶解

    3. 定容至500ml,保持溶液pH为8.0

  2. 配制50XTAE储存液

    1. 称量242gTris base

    2. 溶解在400ml去离子水中

    3. 加入57.1ml冰醋酸,100ml,0.5M EDTA(pH 8.0)

    4. 定容至1L

    5. 常温储存,(此时pH约为8.5)

  3. 配制TAE工作液(40mM Tris醋酸 1mM EDTA)

    1. 稀释50*TAE储存液至1*TAE工作液
