In all, 2.5 × 104 CAR T cells were labeled with 1 nM CellTrace Far Red (Thermo-Fisher) according to the manufacturer’s instructions and co-cultured with CD229-positive U-266 cells and CD229-negative K562 cells in AIM V media for 72 h at 37 °C. Dye dilution as a measure of proliferation was determined using an LSRFortessa flow cytometer (BD).
