Transfection of miRNA into mAG-haESCs

miRNA mimics and inhibitors were synthesized by GenePharma and transfected into mAG-haESCs by using Lipo2000. Briefly, 6 μL of Lipo2000 was added to 400 μL of Opti-MEM, while 10 μL of mimic/inhibitor (20 μM) was added to 400 μL of Opti-MEM. After the mixture was allowed to stand at room temperature for 5 minutes, diluted Lipo2000 was added to the diluted mimics/inhibitor, mixed thoroughly, and allowed to stand at room temperature for 15 minutes. The mixture was added to a 12-well plate of mAG-haESCs at the proper density.
