The sections were dewaxed and hydrated, and endogenous peroxidase was blocked with hydrogen peroxide. Then antigen was retrieved by citrate buffer. After that, the slides were incubated with primary and secondary antibodies, used DAB staining, counterstained with hematoxylin buffer and sealing. Finally, the expression of CD34 in rat breast tissue was observed under Olympus BX53 microscope (Janpan) and analyzed with Image J software.
