The activity of caspase-8, caspase-10, and caspase-12 was assessed by means of a Biovision kit (Biovision, Lyon, France) according to the manufacturer’s instructions. The assay is based on the detection of cleavage of substrate IETD-AFC (AFC: 7-amino-4-trifluoromethyl coumarin) that emits blue light (λmax = 400 nm), upon cleavage of the substrate by FLICE or related caspases, which can be quantified using a fluorometer or a fluorecence microtiter plate reader. Samples were transferred into a 96-well plate, and the fluorescence assessed using a Tecan fluorometer (TECAN Infinite® 200 PRO).
