I would like to know the best way to arrange positive and negative controls, blanks and test samples (cell suspension) in the wells of an Elispot plate. Is it possible to show a template of the layout in your reply. Also, which is the best way to represent repeats of the experiment? Is it better like 1a, 1b, 2a, 2b, etc, or plate A: 1a, 2a, 3a, then plate B: 1b, 2b, etc.
I wish to carry out an Elispot assay for IFN-gamma secretion by T cells, with the hope of using CEF pooled peptide as positive control, and AFP and NY-ESO-1 antigens as liver cancer-specific antigens for stimulating the T cells in the samples. Is this really a good idea?, which is the best place to source these antigens. Thank you.
