The islets were isolated from the female mice as previously described (24, 25). C57BL/6J female mice were anesthetized by intraperitoneal injecting 100 mg/kg pentobarbital (Nembutal, Abbott Laboratories). Then their pancreatic gland was isolated and digested by collagenase. Pancreatic islets were then separated from the digest by density gradient centrifugation using Histopaque 1077. Microdissected, and hand-picking were conducted under a stereomicroscope to ensure high purity.
