The immunoprecipitated YAP (WT or T241A) with protein A/G-Sepharose were incubated with reaction buffer (20 mM HEPES, 50 mM NaCl, 1 µM PuGNAc, and 5 mM MnCl2 with protease and phosphatase inhibitors). Then 2 µl of Gal-T1Y289L (Invitrogen, Carlsbad, CA, USA) and 2 µl of 0.5 mM UDP-GalNAz (Invitrogen) were added. The mix was incubated overnight at 4 °C. After washing twice, the samples were reacted with biotin alkyne (Invitrogen) or tetramethyl-6-carboxyrhodamine (TAMRA) alkyne (Invitrogen) and finally detected by WB using HRP HRP-labeled Streptavidin (Beyotime, #A0303) or antibodies against TAMRA (Invitrogen, #A6397)19.
