Polyacrylamide gel electrophoresis (PAGE) analysis

The reaction mixtures were prepared according to the “PHAmp reaction for target DNA detection” guidelines, except that 500 nM HP was used instead of 25 nM HP and 1X SYBR Green I was excluded. After incubation at 37 °C for 60 min, a 10 μL aliquot of the reaction solution was resolved on a 15% polyacrylamide gel. Electrophoresis was carried out at a constant voltage of 120 V for 120 min using 1X TBE as a running buffer. After staining with ethidium bromide (EtBr) (Bioneer, Daejeon, South Korea), an image of the gel was taken with a Gel Doc EZ Imager (Bio-Rad, CA, USA)23,24,35.
