H295R and SW13 cells were seeded into 48‐well plates at a density of 2 x 104 cells/well, and then transfected with a specific sirt1 siRNA. Additionally, cells were treated with sirtinol (40 μmol/L) and mitotane (10 μmol/L) alone or in combination for 72 hours. Cells were observed under an inverted phase contrast microscope (Olympus CKX53) with a 10X objective.
