Cell migration and invasion were examined by Transwell assays. In Transwell invasion assay, porous membranes were pre-coated with Matrigel (BD Biosciences). A549 and H1299 cells suspended in serum-free medium (200 μL) was transferred to the pre-coated apical chamber. The basolateral chamber was added with 500 μL medium plus 10% FBS. After 24 h of incubation, cells left on the upper side of the membrane were removed. Invasive cells were stained with crystal violet. Cells in the five random fields were counted using a microscope. Migration assay was performed using uncoated apical chambers according to a similar protocol.
