Lung tissues were fixed with glutaraldehyde buffered fixative (3%; pH 7.2) at 25°C for 2-3 days. Tissues were embedded in Spon 812 (SPI-Chem, Inc.; Structure Probe, Inc.) at 60°C for 48 h. Sections (thickness, 60-80 nm) were stained with 2% uranium acetate at 25°C for 10 min and lead citrate at 25°C for 5 min. Following washing with PBS, sections were analyzed using a JEM-2000EX transmission electron microscope (JEOL, Ltd.; magnification, ×17,000) in three randomly selected fields of view.
