Peripheral blood was collected from each patient at the start of the study and 3 months after the final infusion of allogeneic MSCs. Peripheral blood mononuclear cells (PBMCs) were prepared from heparinized blood by Ficoll ± Hypaque (GE Healthcare, PA) density-gradient centrifugation. Cell culture was performed as described previously [21]. In brief, a cell suspension was adjusted to a concentration of 106 cells/mL in RPMI 1640 medium supplemented with 10% fetal calf serum, 100 U/mL penicillin, 100 mg/mL streptomycin, and 2 mM L-glutamine. The cell suspension (1 ml) was dispensed in 24-well multiwell plates (Nunc, Roskilde, Denmark).
