HeLa or T98G cells were seeded in 96-well culture plates and cultured overnight to adhere at 37°C. After pretreatment with helichrysetin at 37°C for 30 min, cells were stimulated with TNF-α (20 ng/ml) for 24 h at 37°C. Subsequently, cells were fixed with 4% paraformaldehyde at room temperature for 10 min and washed with PBS. Cells were incubated with 50 μl Hoechst 33258 staining solution at room temperature for 5 min and then washed twice with PBS. Cell morphology was observed and captured under a fluorescence microscope (magnification, ×200).
