The apoptotic rate of both cell lines was evaluated using an Annexin V/Propidium Iodide (PI) apoptosis detection kit (eBioscience™ Annexin V-FITC Apoptosis Detection kit; Thermo Fisher Scientific, Inc.), according to the manufacturer's instructions. Cells were seeded in a 6-well plate (Corning Inc.) at a concentration of 2×105 cells/ml. Following treatment with light and cisplatinum, cells were harvested, centrifuged (770 × g for 5 min at 4°C), and washed with phosphate-buffered saline (PBS; Euroclone). The cells were then resuspended in binding buffer plus Annexin V following 10 min of incubation at room temperature in the dark. Finally, cells were stained with PI and assessed using the FACSCalibur flow cytometer (BD Biosciences) equipped with Cell Quest software set on a logarithmic scale (BD Biosciences). A minimum of 20,000 cells was acquired for each sample. Data were analyzed using FlowJo™ Software (FlowJo™ Software for Windows version 7.6.1.; FlowJo LLC).
