3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazoliumbromid (MTT) was dissolved to a concentration of 5 g/L in 0.9% NaCl solution and sterile filtered with 0.22 µm filters. 24 h after seeding of 4500 cells per well in 96-well plates in complete α-MEM, incubation with URS at the concentrations listed in Table S1 in α-MEM without FCS or PL was started. 0.1% sodium azide was used a positive control. After 24 h, 10 µl MTT solution per well were added and incubation was continued for 4 h. Wells were washed with PBS and formazan crystals formed by metabolically active cells were dissolved in 100 µl isopropanol. Absorbance was measured in a microplate reader at a wavelength of 590 nm and 690 nm as a reference. After subtraction of background absorbance, means of five replicates were calculated and expressed relative to the appropriate solvent control.
