Gal3-F(ab')2 mAb was produced by pepsin digestion of a well-characterized rat anti-Gal3-mAb (clone M3/38, Mabtech) binding to an amino-terminal common epitope of human and mouse Gal3 21. The digestion was performed after preconditioning of the antibody at acidic pH, as reported earlier 22.

The conjugation with the AlexaFluor488 fluorescent dye and with a Cy5.5-NHS ester derivative, as well as the conjugation to desferrioxamine (DFO) and radiolabeling with 89Z was carried out as previously reported (supplementary material) 20.
