To conduct BiFC assays, the ORF of SmMYB1 fused to cYFP and the ORF of SmMYC2 fused to nYFP were cloned into pXY104 and pXY106 vectors, respectively (Supplementary Fig. S5). The resulting constructs were introduced into A. tumefaciens strain GV3101, which were then infiltrated into 6-week-old N. benthamiana leaves using a 1 mL needleless syringe, as described previously40. The infected leaves were examined 48 h after infiltration. YFP fluorescent signals were captured using a confocal spectral microscopy imaging system (Leica TCS, SP5).
