The complete ORF of SmMYB1 was cloned into the SpeI and BstEII restriction enzyme sites of pCAMBIA2300+ under the control of the CaMV 35S promoter and NOS terminator to generate a pCAMBIA2300+SmMYB1 construct (Supplementary Fig. S2). The DNA sequence encoding SRDX (LDLDLELRLGFA) was inserted into the ORF of SmMYB1 before the stop codon (TAA)26. This sequence was then subcloned into the SpeI and BstEII sites of a pCAMBIA2300+ vector to generate a pCAMBIA2300+-SmMYB1-SRDX construct (Supplementary Fig. S2). All the constructs were introduced into Agrobacterium strain C58C1, which were then transformed into S. miltiorrhiza to produce transgenic lines with hairy roots, as described previously5,38.
