Heart graft tissue was removed and fixed in 10% formalin for 48 hours prior to transfer to 70% ethanol for storage. Tissue was embedded in paraffin, sections were cut and stained by H&E, and immunohistochemistry stains for CD4 (catalog 14-9766-80, Invitrogen) and CD8 (catalog 14-0808-82, Invitrogen) were performed. Slides were then scanned using the CRI Panoramic Whole Slide Scanner (PerkinElmer). Grafts were scored in a single blind manner on a 5-point scale, with 0 to 5 points given for gross histopathological abnormalities, scarring, decellularization, and extent of mononuclear cell infiltration.
