Each group (5-8F/DDP, 5-8F/R and the control 5-8/F) cells were seeded in 96-well plates at a density of 2 × 103 per well, and cultured at 37°C in 5% CO2 for the indicated time (24, 48, 72 and 96 h). At the different time points, cells were treated with DDP at different concentration (0.5, 1, 2 and 4 μg/ml) or X-irradiation at different dose (2, 4, 6 and 8 Gy), respectively. After treatment, the cells were incubated with 20 µl 5 mg/ml 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) solution reagent (Sigma-Aldrich Corp.) for 4 h, and then the cell viability was evaluated by MTT assay. The absorbance at 490 nm (A490) was measured using a microplate reader (Bio-Tek Inc., Winooski, VT, USA).
