Passage 1 AECs were seeded at about 80% confluence in four-well coverslips and incubated at 37°C with 5% CO2 for 24 hours. AECs were then serum-starved for 1 hour, rinsed with sterile 1× PBS, and then scratched evenly down the middle of the well, in a straight line, with a 200-μl pipette tip (7). Cells were washed with sterile 1× PBS four times to remove any cell debris. Cell were then treated with TGF-β (15 ng/ml) or P4 (30ng/ml) for 1, 24, and 38 hours. Bright-field and confocal microscopy documented wound closure, morphology, and vimentin/CK-18 staining.
