Primary hepatocytes were infected 4 to 6 hours after plating with adenoviruses expressing either enhanced green fluorescent protein (EGFP) (Ad-EGFP) or a constitutively active form of PKD3 (Ad-mycPKD3ca) at an multiplicity of infection (MOI) of 10. Medium was replaced the following morning, and cells were used for experiments 36 to 48 hours after infection. Transduction efficiency, which was 100%, was assessed by analyzing the expression of the EGFP reporter (which was present in all adenoviruses).
