FCS was measured using a 40× objective (NA, 1.2) on a Zeiss LSM 780. A diffraction-limited spot was focused on the basal or apical plane of the cells, and three measurements for 10 s each were taken on each point. We measured a minimum of 10 cells for each condition. The autocorrelation functions were fitted with a 2D + Triplet model to obtain the transit diffusion time and the number of particles.
