Lentiviruses were produced by cotransfecting MISSION lentiviral packaging mix (Sigma) into 293T cells using calcium phosphate. Ecotropic p110α retrovirus was produced by calcium phosphate transfection of pBabePuro (p110αH1047) into Phoenix-Eco cells. At 48 to 72 hours after transfection, viral supernatants were collected and filtered. Viral supernatants were then either stored at −80°C or used immediately to infect cells in combination with polybrene at 8 μg/ml. MEFs were selected with puromycin (4 μg/ml; Invitrogen) or blasticidin (2.5 μg/ml; Invitrogen). YAPC and H358 cells were selected with puromycin (6 μg/ml).
