Mock- and TB40/E-5–infected PBMCs were harvested after infection as detailed above and resuspended in PBS with 1% hiFBS. Following incubation on ice for 15 min in the presence of human Fc block (1:100; eBioscience), PBMCs were stained for 30 min on ice with CD36APC (1:100; BioLegend) or CD14APC (1:100; BioLegend), washed in PBS two times, and subsequently stained with calceinBV421 (1:500) for 15 min at room temperature, washed two times, and then analyzed with BD Fusion flow cytometer and FlowJo 10.1 software. All biological and technical replicates were included in analysis.
