HEK293T cells were transfected with calcium phosphate with the plasmids listed in table S6. In brief, 10 μg/ml of each plasmid and 75 μl of 2.5 M CaCl2 were added to deionized water to a final volume of 750 μl. Seven hundred fifty microliters of 2× HEPES buffered saline (HBS) [50 mM HEPES (pH 7.05), 280 mM NaCl, and 1.5 mM Na2PO4] was added dropwise while aerating the mixture. Solution was incubated for 5 min at room temperature before the dropwise addition to a 10-cm plate with 30 to 40% confluent HEK293T cells. Six hours after transfection, the medium was changed to prewarmed DMEM +10% FBS. Experiments were performed 14 hours after transfection.
