Total IFN secreted by pDCs was titrated on STING-37 reporter cells, which correspond to HEK293 cells stably expressing an IFN-stimulated response element (ISRE)–luciferase reporter gene (54). A standard curve was established by applying known titers of recombinant human IFN-α2a (R&D Systems) onto STING-37 cells. Luciferase induction in STING-37 cells was determined using the Bright-Glo reagent (Promega), according to the manufacturer’s recommendations. Quantification of individual IFN subtypes (IFN-α2, IFN-β, IFN-γ, IFN-λ1, and IFN-λ2/3) was performed using the LEGENDplex Kit from BioLegend (human antivirus response panel), according to the manufacturer’s recommendations.
