qPCR to determine the AAV genome copy number was performed using genomic DNA extracted from the retina or RPE. After measuring the genomic DNA concentration using a Quantus fluorometer together with the Quantifluor Dye system (Promega), 50 ng of genomic DNA was subjected to qPCR analysis using an AAVpro titration kit (Takara). Thermocycling conditions were as follows: 95°C, 2 min followed by 35 cycles of 95°C, 5 s; 60°C, 30 s. The number of AAV genome copies was calculated against a standard curve. The number of diploid mouse cells was calculated using a conversion factor of ~1.6 × 104 diploid cells per 100 μg of genomic DNA.
